Description

Principle of the assay: This assay employs a two-site sandwich ELISA to quantitative MUC5AC in Human serum, plasma. An antibody specific for MUC5AC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MUC5AC present is bound by the immobilized antibody. After removing any unbound substances, a biotin - conjugated antibody specific for MUC5AC is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC5AC bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: MUC5AC is a secreted mucin aberrantly expressed by polypoid colorectal adenomas. It has been hypothesised that the "normal" surrounding colorectal mucosa expresses MUC5AC as a field change phenomenon that can be used to predict adenoma recurrence following resection.Mucins are high molecular mass, highly glycosylated macromolecules that are the major components of mucus secretions. MUC5B is a salivary mucin that is thought to contribute to the lubricating and viscoelastic properties of whole saliva. It is composed of 14.9% protein, 78.1% carbohydrate, and 7% sulfate.
